18F-Fallypride的比活度对多巴胺D2受体显像的影响

摘要:

采用亲核反应制备18F-Fallypride。将不同浓度的19F-Fallypride与18F-Fallypride混匀模拟比活度变化,经尾静脉注射正常小鼠(3.7 MBq)后10 min行Micro PET显像,勾画纹状体为感兴趣区域,计算其摄取18F-Fallypride的变化。结果表明,各组纹状体摄取18F-Fallypride的量随比活度降低而降低,摄取值与19F-Fallypride的浓度对数呈S型曲线变化。18F-Fallypride的比活度不低于1.35 PBq/mol时,可保证18F-Fallypride PET显像的准确性。结果可见,18F-Fallypride比活度是多巴胺D2受体显像的重要影响因素。

Abstract:

To study the specific activity requirement of 18F-Fallypride for dopamine D2 receptor PET imaging, 18F-Fallypride was prepared by nucleophilic reaction and purified by HPLC. The specific activity was measured. Different concentrations of 19F-Fallypride and 18F-Fallypride (3.7 MBq) were mixed to simulate the specific activity. After the mixture was intravenous injected into normal mice 10 min, Micro PET was carried out, and then the uptake of 18F-Fallypride in striatum was analyzed. The striatum uptake of 18F-Fallypride decreased when the specific activity decreased, and the uptake value with the log concentration of 19F-Fallypride exhibited S-shaped curve. Based on these, 18F-Fallypride can be used for striatum Micro PET imaging with the specific activity no less than 1.35 PBq/mol. The specific activity is an important factor in the 18F-Fallypride dopamine D2 receptor imaging.
